1. OverView:
Toremifene citrate is an oral selective estrogen receptor modulator (SERM) which helps oppose the actions of estrogen in the body.Toremifene citrate is FDA-approved for use in advanced (metastatic) breast cancer. It is also being evaluated for prevention of prostate cancer under the brand name Acapodene.
2. Background:
In 2007 the pharmaceutical company GTx, Inc was conducting two different phase 3 clinical trials; First, a pivotal Phase clinical trial for the treatment of serious side effects of androgen deprivation therapy (ADT) (especially vertebral/spine fractures and hot flashes, lipid profile, and gynecomastia) for advanced prostate cancer, and second, a pivotal Phase III clinical trial for the prevention of prostate cancer in high risk men with high grade prostatic intraepithelial neoplasia, or PIN. Results of these trials are expected by first quarter of 2008.
3. Use:
If You Are a Postmenopausal Woman With Metastatic Breast Cancer of Estrogen-Receptor Positive (ER+) or Unknown Tumor, Fareston Provides Another Option
Fareston Is a Selective Estrogen-Receptor Modulator (SERM) That:
1.Is clinically proven. In a number of important measurements of a drug's effectiveness in treating metastatic ER+ breast cancer in postmenopausal women, Fareston worked just as well as tamoxifen.
2May provide the continued benefits of endocrine therapy. Fareston may offer you the continued benefits of endocrine therapy if you cannot tolerate or are otherwise unable to take other endocrine therapies.
3Fareston may be an effective alternative for patients who have stopped taking other endocrine therapies because of joint pain.
4Is available and accessible. Fareston is widely available on commercial and Medicare Part D formularies and is covered by most commercial and Medicare insurance plans.
4. Food and drug interactions
There are no known interactions of toremifene citrate with any food. This drug may interact with other medications. Tell your doctor and pharmacist about all of the medications that you are currently taking.
5.Absorption and Distribution:
1.Absorption:
Well absorbed; absorption is not influenced by food. T max is 3 h. Steady state is reached in approximately 4 to 6 wk.
2.Distribution:
Vd is 580 L. Approximately 99.5% is bound to plasma proteins, mainly albumin. Mean distribution half-life is approximately 4 h.
6. Dosage and Administration:
Breast Cancer--Adults
PO 60 mg once daily with or without food. Treatment is generally continued until disease progression is observed.
